Recent Trials Paint a More Complicated Picture
Docherty et al. (2023) — Pilot RCT (n=41, ages 18–45): Found worse delayed word recall in lion’s mane group vs. placebo. Not a result you’ll find in brand marketing.
Surendran et al. (2025) — Acute crossover RCT (n=18, ages 18–35, 3g of 10:1 extract): No significant improvement in global cognitive function or mood. Only a pegboard fine motor task improved.
Saitsu et al. (2019) — 12-week RCT, healthy adults 50+ (n=31): Improvement on 1 of 3 cognitive tests. Both groups improved, weakening the signal.
The NGF mechanism is scientifically legitimate. Human evidence is mixed, small-scale, and mostly short-duration. The most promising results come from populations with pre-existing cognitive decline. Recent trials in healthy young adults have not shown the benefits that marketing suggests. None of that means the stack is worthless — it means go in calibrated.
